What is phage display technology?
Phage display technology is to insert the DNA sequence of the foreign protein or polypeptide into the appropriate position of the phage coat protein structural gene, so that the foreign gene is expressed along with the expression of the coat protein. At the same time, the foreign protein is renewed with the phage.
Principles of phage display technology
Phage display technology is to clone the polypeptide or protein coding gene or target gene fragment into the appropriate position of the phage coat protein structural gene. Under the condition that the reading frame is correct and does not affect the normal function of other coat proteins, the exogenous polypeptide or protein can be combined with the coat. The protein is expressed in fusion, and the fusion protein is displayed on the surface of the phage as the progeny phage is reassembled. The displayed polypeptide or protein can maintain a relatively independent spatial structure and biological activity to facilitate the recognition and binding of the target molecule.

Affinity maturation (based on phage display and high-throughput screening platform)
- Based on the optimization of the binding kinetics (dissociation rate), the Kd value can be increased to below nM level, or even to pM level
- Both scFv and Fab formats can be used to construct CDR mutation libraries.
- Using biotinylated antigen for liquid library panning, the concentration of the antigen can be lower.
- We use the off-rate screening method to screen pM-level binding molecules, and use non-labeled antigens for competitive binding.
- The enriched clones are amplified in 96-well plates, and high-throughput automated screening equipment is used to rank the affinity of different clones.
- The best clones that have been identified can be converted into full-length IgG and subjected to further Biacore and biological and functional analysis.
